【摘要】:Measurementofbacterialdensitiesisgenerallythefirststepinestablishingarelationshipbetweenbacteriaandotherbiochemicalprocesses.Itisknownthattheclassicalplatecountprocedureunderestimatesbacterialdensitieswhiletheepifluorescencedirectmicroscopicproceduremoreaccuratelydepictsthetotalnumbersofnonviableordormantandviablecellsinawatersample.Theacridine-orangeINT-formazanreductiontechniqueprovidesinformationonthetotalconcentrationsofbacteriaaswellasthatproportionwhichareactivelyrespiringandthusinvolvedindegradativeprocesses.Theacridine-orangeINT-formazanreductiontechniqueisbothquantitativeandprecise.Thisprocedureisidealforenumeratingbothpelagicandepibenthicbacteriainallfreshwaterandmarineenvironments.Theprocesscanbeemployedinsurveystudiestocharacterizethebacteriologicaldensitiesandactivitiesofenvironmentalwaters.1.1Thistestmethodcoversthedetectionandenumerationofaquaticbacteriabytheuseofanacridine-orangeepifluorescencedirect-microscopiccountingprocedure.Thistestmethodisapplicabletoenvironmentalwatersandpotablewaters.1.2Certaintypesofdebrisandothermicroorganismsmayfluoresceinacridine-orangestainedsmears.1.3Theproceduredescribedrequiresatrainedmicrobiologistortechnicianwhoiscapableofdistinguishingbacteriafromotherfluorescingbodiesonthebasisofmorphologywhenviewedathighermagnifications.1.4Useofbrightlightpermitsdifferentiationofsinglebacteriawherereducedformazanisdepositedatthepolarends.1.5Approximately104cells/mLarerequiredfordetectionbythistestmethod.1.6Minimalcellsizewhichallowsthedetectionofformazandepositsisrepresentedbybacteriaof0.4x03BC;m.21.7ThevaluesstatedinSIunitsaretoberegardedasstandard.Nootherunitsofmeasurementareincludedinthisstandard.1.8Thisstandarddoesnotpurporttoaddressthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
